???metadata.dc.description.resumo???: O uso de ansiolíticos tem crescido nos últimos anos no mundo todo, pois um dos problemas mais recorrentes da população mundial é a ansiedade e distúrbios do sono. O desenvolvimento de novos sistemas de liberação de fármacos torna-se cada vez mais importante na promoção das ações terapêuticas a fim de reduzir as limitações nas terapias existentes. Diante disso, o uso de polímeros como a quitosana, que apresentam características de biodegradabilidade, biocompatibilidade, mucoadesão, associada com outros polímeros como a gelatina, apresenta-se como uma alternativa viável para obtenção de um sistema de liberação controlada de diversos fármacos, incluindo os benzodiazepínicos. Desta forma, o presente trabalho teve como objetivo obter filmes de Quitosana/Gelatina para liberação do Midazolam. Os filmes foram preparados pelo método de evaporação de solvente, através da dissolução da quitosana em ácido acético (1% v/v), para uma concentração final da solução polimérica (1% m/v), seguida da incorporação de gelatina (20% m/m) e Midazolam (0,03mg/mL). Após a obtenção os filmes foram caracterizados utilizando a MO, DRX, FTIR, Grau de Intumescimento, Biodegradação e Molhabilidade por Medida de Ângulo de Contato. Na MO observou-se no material superfícies uniformes e densas. No ensaio de DRX pode-se confirmar que a incorporação do fármaco e da gelatina na solução de quitosana acarretou na redução da cristalinidade do filme. No ensaio de FTIR evidenciou-se a interação do fármaco e da gelatina com os grupamentos hidroxila e amina da quitosana. Observou-se um aumento do grau de intumescimento com a adição do fármaco. Na análise de biodegradação, observou-se que a mesma é influenciada pela adição de gelatina e principalmente pelo fármaco. Os resultados de Molhabilidade demonstraram que o material tem caráter hidrofílico e que a incorporação do fármaco possibilitou o aumento da hidrofilicidade do mesmo. Mediante o exposto nota-se que o sistema apresentou homogeneidade, adequada interação química entre os constituintes e foi possível obter filmes de Quitosana/Gelatina incorporadas com Midazolam. Sendo assim, conclui-se que os filmes apresentam potencial para utilização em sistema de liberação de midazolam, vislumbrando o uso como biomaterial biodegradável com ação ansiolítica.
Abstract: The use of anxiolytics has grown in recent years worldwide, as one of the most recurrent problems in the world population is anxiety and sleep disorders. The development of new drug delivery systems becomes increasingly important in promoting therapeutic actions in order to reduce the limitations in existing therapies. Therefore, the use of polymers such as chitosan, which exhibit characteristics of biodegradability, biocompatibility, mucoadhesion, associated with other polymers such as gelatine, is presented as a viable alternative to obtain a controlled release system of several drugs, including benzodiazepines. Thus, the present work aimed to obtain Chitosan / Gelatin films for the release of Midazolam. The films were prepared by the solvent evaporation method, by dissolving the chitosan in acetic acid (1% v / v), to a final concentration of the polymer solution (1% m / v), followed by the incorporation of gelatin (20% m / m) and Midazolam (0.03mg / ml). After obtaining the films were characterized using MO, DRX, FTIR, Degree of Swelling, Biodegradation and Wettability by Contact Angle Measurement. In OM, uniform and dense surfaces were observed in the material. In the XRD assay it can be confirmed that the incorporation of the drug and gelatin in the chitosan solution led to the reduction of film crystallinity. In the FTIR assay the interaction of the drug and gelatin with the hydroxyl and amine groups of the chitosan was evidenced. As to the degree of swelling, a greater increase of the samples with the drug was observed. In the biodegradation analysis, it was observed that it is influenced by the addition of gelatin and mainly by the drug. The Wettability results showed that the material has a hydrophilic character and that the incorporation of the drug enabled the increase of the hydrophilicity of the same. It was observed that the system presented homogeneity, adequate chemical interaction between the constituents and it was possible to obtain Chitosan / Gelatin films incorporated with Midazolam. Thus, it is concluded that the films present potential for use in the midazolam release system, with the use of biodegradable biomaterial with anxiolytic action.
