Abstract:
Due to the increased incidence of fungal infections, especially Candida albicans, and
the increasingly frequent reports of fungal resistance, it is necessary to search for
new treatment options. Natural products represent a vast field of research, with
many reports in the literature of substances of natural origin with potential
antifungal, among these substances are essential oils and their components. This
study aims to investigate the activity of citronellol and geraniol against C. albicans
in order that these substances are components of some essential oils that have
antimicrobial activity. Through the microdilution technique was determined the
Minimum Inhibitory Concentration (MIC) of geraniol and citronellol; being that
when an aliquot from each well of the microtiter plate where no growth was plated
on Sabouraud Dextrose Agar (SDA) met the minimum fungicidal concentration (
CFM); further by microdilution technique, we investigated the ability of substances
interacting with the ergosterol and interfering with the cell wall. The results showed
that these substances have antifungal activity, the MIC of geraniol and citronellol
was 250 mg / mL for all strains tested. The CFM of geraniol against strains ICB-12
and ATCC 76615 was 1000 mg / mL, against all other strains was 250 mg / mL. The
CFM of citronellol against ICB-12 strain was 500 mg / mL, against the other strains
was 250 g / mL.Na investigation of the mechanism of action was found that both
geraniol as citronellol ligated to ergosterol, which can be observed by increasing the
MIC. There was no increase in MIC of either oils in the presence of sorbitol, which
suggests that the action of geraniol and citronellol not occur per share in the cell
wall. The results show that these substances are potential antifungal agents, with
strong antimicrobial activity, being proven by their considerable values of MIC and
MFC, as to its mechanism of action by its binding to ergosterol, draw a profile of a
possible target that offers selectivity in the action of these substances may be
continued studies to detailed design of your facility as well as conducting preclinical
studies clinicians.
