Estrogen replacement therapy for menopausal women may be administered systemically or vaginally, the latter being the optimal route of administration for urogenital atrophy. Polymer rings for hormone release become a candidate for treatment, however, it is necessary to remove them after their period of functionality is 21 days. Therefore, the use of polymers such as chitosan, which has characteristics of biodegradability, biocompatibility, mucoadhesion presents as an alternative for the development of the rings. Thus, the present work aimed the development of biodegradable rings for intravaginal release of promestriene. The rings were developed by dissolving the chitosan in lactic acid (1% v / v), to a final concentration of the polymer solution (5% m / v), followed by the incorporation of gelatin (30% w / w) and 10% mg / g of promestriene concentration is used in commercial rings in 30 mL of chitosan solution to produce a ring. A ring-shaped template was used and the chitosan / gelatin / promestriene solution was coagulated in 1M sodium hydroxide, then the samples were washed and submerged in polyethylene glycol (PEG) 400 for 24 h. The samples were then submitted to drying and characterized by optical microscopy (OM), Fourier Transform Infrared Spectroscopy (FTIR), Wettability by contact angle measurement and mechanical tensile testing. In the OM the samples were dense, resembling the commercial ring sample. The FTIR results demonstrated that the drug interacted chemically with the polymer matrix, possibly through hydrogen bonds, dipole-dipole bonds and induced dipole. The mechanical tensile test showed that the incorporation of the drug made possible the increase in resistance. In the wettability by measurement of contact angle, it was verified that all the samples showed contact angle equal to 0 ° characterizing them as hydrophilic. An increase in the degree of swelling with the addition of the drug was observed. In the biodegradation analysis, it was observed that it is influenced by the addition of gelatin and mainly by the drug. It was concluded that the prepared samples presented similarities with the commercial and appropriate chemical interaction between the constituents, through the methodology and
processing techniques used it was possible to develop a chitosan/gelatin/promestriene
ring.
