Resumo:
The purpose of this work was to evaluate the best source tissue and the most efficient DNA
extraction method for the diagnosis of canine visceral leishmaniasis by PCR. Samples were
taken from spleen, liver, popliteal lymph node, bone marrow, conjunctiva and peripheral
blood of naturally infected dogs, from which 16 were asymptomatic and 2 symptomatic. DNA
extraction was performed with commercial kits and isopropanol or sílica methods. The results
suggest that the best association includes the extraction of DNA from popliteal lymph node
using a kit. In the conditions described here, the PCR sensitivity reached 72% in a sample
where 88% of the dogs were asymptomatics.
