Resumen:
Stingless bees have been victims of fragmentation in their natural habitats.
The conservation of these species and their sustainable management is
important to maintaining the Caatingaecosystem. In order to initiate studies of
genetic variability in populations of native bees, this work aim to evaluate
different methods of DNA extraction from thorax and wings ofF.doederleini, for
being used in molecular phylogeny. The samples were collected in Santana do
Seridó / RN and four protocols were tested: 1: Brazol ® reagent, dehydrated
and frozen bees and 0.2% 2-mercaptoethanol in the lysis buffer; Protocol 2:
Brazol ® reagent, dehydrated and frozen bees further 1μL of 2-mercaptoethanol
into the samples; Protocol 3: fractionated silica, dehydrated and frozen bees,
0.2% 2-mercaptoethanol in the lysis buffer; Protocol 4: fractionated silica,
dehydrated and frozen bees, further 2μL of 2-mercaptoethanol into thesamples.
DNA extracted from dehydrated and frozen bees with Brazol ® was
quantitatively and qualitatively superior to that obtained with silica.
