Effect of juliprosopine on mitochondrial bioenergetics. The objective of this work was to evaluate the effect of juliprosopina, an alkaloid from Prosopis juliflora, common plant from Brazilian Northeast that causes outbreaks in cattle using rat liver mitochondria for bioenergetics tests. Thirty kilogram of Prosopis juliflora pods were collected in Patos – PB and the juliprosopina were isolated at Laboratório de Tecnologia Farmacêutica (LTF) from UFPB where a total alkaloid fraction went through a Sephadex column and fractions were united and put in a silica card 20X20 cm for extraction. The main compound was identified by Nuclear Magnetic Resonance H1 and C13. Mitochondrial bioenergetics was assessed at Bioquemistry Laboratory at UNESP/Dracena-SP. Male, 200g, Wistar rats were used for mitochondrial isolation. The animals were euthanized by decapitation under ethylic ether anesthesia. Rat liver were taken, homogenized and centrifuged under different speeds. Mitochondrial respiration, membrane potential and ATP synthesis were assessed. Juliprosopine was tested at 5, 10, 25 and 50 μM. The alkaloid raised oxygen consumption at state IV of mitochondrial respiration in 25 μM. Membrane potential was decreased at same concentration. The alkaloids inhibit ATP synthesis in 10 μM concentration. Results show that juliprosopine uncouples and decreases ATP synthesis which might be connected to its toxicity.
