Mangabeira (Hancornia speciosa Gomes) fruit species native of Brazil has economic potential for exploitation of drugs or fruit production. However, the species has a low germination rate and recalcitrant seeds. In this context, the objective of this study was to test the effect of proline modify the step of pre-cultivation of lateral gems before cryopreservation. The gems were extracted from shoots grown in vitro mangabeira and pre-grown in WPM culture medium supplemented with 0.3 M sucrose combined with different concentrations of proline (0.0, 0.1, 0.2 and 0.3 M) and for different times (24 and 48 hours). After the pre-cultivation the shoots were treated with PVS2 at 0 °C for 15 minutes and cryopreserved by vitrification droplet. After cryopreservation, the samples were reheated and gems cultivated in WPM containing 0.2 μM BAP in a growth chamber with a 16h photoperiod. After 30 days of culture were evaluated survival and regeneration of shoots by Skott-Knott test (p ≤ 0.05). The preculture in 0.3 M sucrose + 0.1 M proline for 24 hours caused a significant increase in the percentage of survival (83.3%) and regeneration of shoots (93.3%).
