Resumo:
The aim this study was evaluated the effect of two induction strategies (pulse and gradual addition) with IPTG in the expression of 503 antigen of Leishmania infatumchagasi in E. coli. In the test batch, IPTG was added to the culture at a final concentration of 1 mM. In fed-batch, 0.2 mM of the inducer was added every hour, reaching a final concentration of 0.8 mM. The procedure of induction by pulses of IPTG indicated an increase of approximately 49% in biomass production compared to batch. The results showedthat the strategy ofinduction using pulses of IPTG (fed- batch) was more effective for 503 antigen expression when compared to the simple batch strategy. Efficacy of the induction strategy was confirmed by electrophoresis.
