The class of proton pump inhibitors (PPIs), omeprazole, lansoprazole, pantoprazole, rabeprazole, dexlansoprazole and esomeprazole, omeprazole being the most prescribed because of the lower cost and easier access. It is widely used in gastrointestinal diseases and acts by inhibiting the enzyme K+/ H+-ATPase responsible for the secretion of gastric acid. Although it is marketed as an effective, safe and inexpensive drug, unnecessary and prolonged exposure can cause harm to users' health. In view of the above, this work had the objective of reviewing the literature on the misuse of omeprazole. The study was made available through the internet and in the library of the Federal University of Campina Grande, Campus de Cuité - PB (UFCG). Data search was done in the Medline, Pubmed, Lilacs, Scielo, Academic and national health committees, using the following terms isolated or in combination: irrational use, omeprazole, proton pump inhibitor, effects adverse reactions, interactions and prescription. We included studies on the proposed theme, published in the last eight years (2010-2018), in the English, Portuguese and Spanish languages and in the format of articles, dissertations, monographs, books and health committees. Based on the results found, the study warns of the undesirable risks and possible interactions that the user is exposed to by making inappropriate, unnecessary and chronic use of omeprazole. There is a need for measures aimed at the rational use of the drug for therapeutic success.
