Abstract:
Various molecular techniques for microbiological analyses have recently been developed, and this has led to the acquisition in large amounts of genetic information about microorganisms found in a given environment, especially facilitated by the use of molecular markers from a DNA sequence. Concurrently with the development based in microbiological cultivation, these molecular biology techniques is now considered as the main study subject for certain fields of experimental biology, due to the technology used which allows to reveal the DNA variability and detect the differences among individuals and consequently among the risks in their pathogenicity. Basic studies on DNA extraction techniques are extremely important for the success of scientific papers in molecular biology field, so, this study aimed to evaluate, by DGGE the genetic variability of water microorganisms from Várzea Grande reservoir in the city of Picuí-PB, applying the CTAB solution for DNA extraction and further analysis. It was possible to observe that the extractions performed in this study have demonstrated a sufficient and high quality DNA yield for the intended analyses and also for future specific genetic analyses. The results obtained with these extractions did not diverge from the standards expected for this type of sample. Therefore, we conclude that the process adopted for the intended analysis was sufficient for obtaining a high quality genomic DNA. The DGGE technique showed to be able for future prospection about the microbiological components from the sample place analyzed. This can be answer how the microbiological community is working in this water environment and how they can influence the life quality of population living around there.
